The reason why most of us think that the abbreviation 'no.' should be written as 'nu.' is that the second letter of 'number' is 'u'. But as all of us know that English borrows many of it's words from many other languages, for example, french, italian, german. So, the word 'number' in short form written as 'no.' for the same reason why ounce is written as 'oz' and pound is written as 'lb' and many such abbreviations. In this case, the abbreviation "no." for the word "number" comes from the numero symbol, № (also written as "No." or "no."), which is in turn derived from Latin numero. As you can see, it has an "o" in it.
